Dating back from 2000's (seems so long ago) there has been very very very many movies some of them have been fantastic and some have just been... well horrible.
Well with this list it is the TOP 5 Comedy Movies since 2000.
5.) Super Troopers
-This movie had so many punch-lines and is just a amazing comedy movie.
4.) Jay and Silent Bob Strike Back
- If you have ever seen this movie, you will know why this hands down ranks in the top 5 funniest movies of the 2000's.
3.) *******
-Need I say more?
2.) Zoolander
-Ben Stillers funniest role that I believe
1.) Team America World Police
-America... _______ Yeah
Other Possibilities:
Hot Fuzz, Shaun of the Dead, Wedding Crashers, Anchorman, Road Trip, South Park The Movie, Clerks II, Club Dread, I can go on and on so I will stop

I am assuming you mean comedies that were made from 2000-present. So I'll include as such:

5.) Shrek
This movie was a surprise when it came out. Looked like just another kid's movie, but was laden with adult level jokes and references. What made it even more classic was all the spoofs on old stories and nursery rhymes. If you knew a story growing up, chances where it was in there somewhere being funny. The ONLY movie that I have ever seen in a movie theatre more than twice; in this case 5 times.

4.) Napoleon Dynamite
This movie wasn't funny to me at all the first time. However I saw it a few more times, and then I finally got it. It's all about how akward everything seems to be, and everything is just so subtle yet blatant. At that point it was absolutely hilarious. It definitly grows on you. It is so out of the norm, and it really stood forth. Plus who couldn't love the dance scene?

3.) National Lampoon's: Van Wilder
This movie gives you so much everytime you see it. So many great one liners, and the main character has the perfect personality for the character. The perfect college movie, but flat out funny either way. Although there was one really gross part, all I have to say is "The bakery is closed".

2.) Anchorman: The Legend of Ron Burgundy
Quite possibly Will Ferrils best movie. The movie is loaded with quotes and scenes that people repeat on a daily basis. Great cast that really brings to life each character. I especially love Brick, player by Steve Carrel. I LOVE LAMP! are you just saying that, or do you really love the lamp?

1.) Supertroopers
Most will say I went out on a limb here, but I think this movie was closest to the best comdey this decade. The movie was so strange and completely unheralded, but did amazing. The jokes were all over the spectrum, from dirty to one lines to akward. Whatever your taste, it seemed to be there. The movie grabs you from the first scene where the cops totally screw with the stoned kids. I could go on and on, but there are so many great moments. Farva... Litering and... the Meow game... and the repeater (and the repeater). So just a great great comedy, and if you haven't seen it; go now and rent it.

Note: This list was EXTREMELY hard for me to make. I tried to select movies that had lasting effect and tag lines. Others considered were (no particular order):

Austin Powers 3, Jay and Silent Bob: Strike Back, Dodgeball, Borat, Oldschool, Waiting, Out Cold, Meet the Parents, Team America, Shaun of the Dead, Saving Silverman, and Blades of Glory.
